I think I've fallen for a colleague but I don't know if she's a lesbian or not. I also don't know if she likes me or not. So, my question is whether it is advisable for me to send her an audio song "can't help falling in love" first then decide whether to tell her I'm interested in her.

I feel like that particular song is a bit too forward. You don't want to make her uncomfortable. You should probably find out her sexual orientation first and come out to her if you haven't already.
